Seventeen years of Shiki Tohno returned to his home after an eight-year absence. Once this decision was taken by his father - a hard and uncompromising head of a long line of Tono, and overturned the expulsion of his sister Akiha, who became his father's death the new leader of the clan.
Although Shiki, who grew up in the care of his uncle with an aunt, a soft and sensitive young man, accustomed to a quiet life, even he is surprised strict rules, introduced his sister, that are more suitable for a besieged fortress, a mansion than an influential family. However, the newly found understanding with a longtime childhood friend - the maids Hisui and Kohaku - helps to defeat Shiki curfew and enjoy the modest pleasures of a normal Japanese guy.

Shiki Tohno
His real surname is Nanaya, and he is the eldest son of the Nanaya clan, a family of powerful demon-slaying assassins, it is revealed in Kagetsu Tohya that he is the son of Nanaya Kiri. His personality is one of that who is slightly mean to those he loves. He is also a very, very heavy sleeper, to such a point that Hisui is unable to awaken him.
Arcueid Brunestud
She is the eponymous Tsukihime (Moon Princess), as evident by the title of her "True Ending." She also has important roles in Kagetsu Tohya and the story mode of Melty Blood. Her behavior is almost child-like, due to the lonely nature of her up bringing, although she is over 800 years old, she spent most of it asleep and knows nearly nothing about the world or how to interact with others.
Ciel
She is a member of the Burial Agency, a secret organization within the Catholic Church made up of skilled assassins, feared even within the Church itself, known for showing no mercy and willing to do whatever it takes to get their task done even to the point of using forbidden magics and creatures.
Akiha Tohno
She is very shy about her feelings, even acting in complete contradiction to them on occasion. Upon the death of their father eight years later, Akiha became the head of the Tohno household and, ignoring protests from the remaining relatives, called Shiki back to live at the family mansion with her.
Hisui
She is responsible for all of the cleaning in the mansion. She refuses to accept help from Shiki with her chores, or to talk with him less formally. She personally wakes him up every morning and waits at the front entrance for him to return from school.
Kohaku
She is very cheerful and upbeat, in contrast to Hisui's stoic demeanor. She is Hisui's twin sister and Akiha's personal servant.
Satsuki "Sacchin" Yumizuka
She plays somewhat different roles in the game and the anime. In the anime, she is portrayed as somewhat more outgoing and a mutual friend of both Shiki and Inui Arihiko, and confesses to Shiki near the end of the series.
Nrvnqsr "Nero" Chaos
He is a man who became a Dead Apostle vampire through magical research. In order to prolong his immortality, he took the bodies of many different types of beasts and made them a part of himself.
Aoko Aozaki
She provides him with the special glasses he has to block his Mystic Eyes of Death Perception ability. She then disappears. She shows up in the epilogue to Tsukihime, where she tells Shiki he's become a fine young man and Shiki finally thanks her for being there for him.
Len
She has a shy personality, but is known to like to play tricks when she is bored. She's the star of Kagetsu Tohya, although this isn't apparent at first. She appears either as a small girl with elf ears or a black kitten. Her role in the original Tsukihime is to give Shiki an erotic dream as a reward for him destroying Nero.
